Actions of sex steroids on kisspeptin expression and other reproduction-related genes in the brain of the teleost fish European sea bass ArchiMer
Alvarado, M. V.; Servili, Arianna; Moles, G.; Gueguen, M.m.; Carrillo, M.; Kah, O.; Felip, A..
Kisspeptins are well known as mediators of the coordinated communication between the brain-pituitary axis and the gonads in many vertebrates. To test the hypothesis that gonadal steroids regulate kiss1 and kiss2 mRNA expression in European sea bass (a teleost fish), we examined the brains of gonad-intact (control) and castrated animals, as well as castrated males (GDX) and ovariectomized females (OVX) that received testosterone (T) and estradiol (E2) replacement, respectively, during recrudescence. In GDX males, low expression of kiss1 mRNA is observed by in situ hybridization in the caudal hypothalamus (CH) and the mediobasal hypothalamus (MBH), although hypothalamic changes in kiss1 mRNA levels were not statistically different among the groups, as...

Ocean warming, eutrophication and consequent decrease in oxygen lead to smaller average fish size. Although such responses are well-known in an evolutionary context, involving multiple generations, it appears to be incompatible with current rapid environmental change. Rather, phenotypic plasticity could provide a means for marine fish to cope with rapid environmental changes. However, little is known about the mechanisms underlying plastic responses to environmental conditions that favour small phenotypes. Our aim was to investigate how and why European sea bass that had experienced a short episode of moderate hypoxia during their larval stage subsequently exhibited a growth depression at the juvenile stage compared to the control group. We examined...

Calcium response of KCl-excited populations of ventricular myocytes from the European sea bass (Dicentrarchus labrax): a promising approach to integrate cell-to-cell heterogeneity in studying the cellular basis of fish cardiac performance ArchiMer
Ollivier, Helene; Marchant, James; Le Bayon, Nicolas; Servili, Arianna; Claireaux, Guy.
Climate change challenges the capacity of fishes to thrive in their habitat. However, through phenotypic diversity, they demonstrate remarkable resilience to deteriorating conditions. In fish populations, inter-individual variation in a number of fitness-determining physiological traits, including cardiac performance, is classically observed. Information about the cellular bases of inter-individual variability in cardiac performance is scarce including the possible contribution of excitation-contraction (EC) coupling. This study aimed at providing insight into EC coupling-related Ca2+ response and thermal plasticity in the European sea bass (Dicentrarchus labrax). A cell population approach was used to lay the methodological basis for identifying the...

Climate change impacts on fish reproduction are mediated at multiple levels of the brain-pituitary-gonad axis ArchiMer
Servili, Arianna; Canario, Adelino V.m.; Mouchel, Olivier; Muñoz-cueto, José Antonio.
Anthropogenic emissions of carbon dioxide in the atmosphere have generated rapid variations in atmospheric composition which drives major climate changes. Climate change related effects include changes in physico-chemical proprieties of sea and freshwater, such as variations in water temperature, salinity, pH/PCO2 and oxygen content, which can impact fish critical physiological functions including reproduction. In this context, the main aim of the present review is to discuss how climate change related effects (variation in water temperature and salinity, increases in duration and frequency of hypoxia events, water acidification) would impact reproduction by affecting the neuroendocrine axis (brain-pituitary-gonad axis). Variations in temperature and...

Contribution of a bioenergetics model to investigate the growth and survival of European seabass in the Bay of Biscay – English Channel area ArchiMer
Dambrine, Chloe; Huret, Martin; Woillez, Mathieu; Pecquerie, Laure; Allal, Francois; Servili, Arianna; De Pontual, Helene.
The European seabass (Dicentrarchus labrax) is a species of particular ecological and economic importance. Stock assessments have recently revealed the worrying state of the “Northern stock”, probably due to overfishing and a series of poor recruitments. The extent to which these poor recruitments are due to environmental variability is difficult to assess, as the processes driving the seabass life cycle are poorly known. Here we investigate how food availability and temperature may affect the growth and survival of wild seabass at the individual scale. To this end, we developed a bioenergetics model based on the Dynamic Energy Budget (DEB) theory. We applied it to seabass population of the Northeast Atlantic region (Bay of Biscay – English Channel area)...

Early exposure to chronic hypoxia induces short- and long-term regulation of hemoglobin gene expression in European sea bass (Dicentrarchus labrax) ArchiMer
Cadiz, Laura; Servili, Arianna; Quazuguel, Patrick; Madec, Lauriane; Zambonino, Jose-luis; Mazurais, David.
European sea bass (Dicentrarchus labrax) inhabits coastal waters and may be exposed to hypoxia at different life stages, requiring physiological and behavioral adaptation. In the present study, we attempted to determine whether regulation of hemoglobin (Hb) gene expression plays a role in the physiological response to chronic moderate hypoxia in whole larvae and hematopoietic tissues (head kidney and spleen) of juveniles. We also tested the hypothesis that hypoxia exposure at the larval stage could induce a long-term effect on the regulation of Hb gene expression. For this purpose, D. labrax were exposed to a non-lethal hypoxic condition (40% air saturation) at the larval stage from 28 to 50 days post-hatching (dph) and/or at the juvenile stage from 196 to...

Genomic organization and spatio-temporal expression of the hemoglobin genes in European sea bass (Dicentrarchus labrax) ArchiMer
Cadiz, L.; Desmarais, E.; Servili, Arianna; Quazuguel, Patrick; Madec, Lauriane; Huelvan, Christine; Andersen, O.; Zambonino-infante, Jose-luis; Mazurais, David.
Hemoglobins (Hb) play a critical role in satisfying the oxygen demand of vertebrate aerobic metabolism. The present study reports the characterization of the European sea bass (Dicentrarchus labrax) Hb genes, including genomic organization, phylogeny, and spatio-temporal gene expression. These Hb genes are divided into two unlinked clusters, the “MN” cluster containing eleven genes (five Hbα genes named MN-Hbα1-5 and six Hbβ genes named MN-Hbβ1–6) and the “LA” cluster consisting of three genes (two Hbα genes named LA-Hbα1-2 and one Hbβ gene named LA-Hbβ1). Comparative analysis of Hb amino acid sequences indicates that most of the important amino acid residues involved in hemoglobin-oxygen binding, particularly in the Bohr and Root effects, are generally...

Long-term exposure to near-future ocean acidification does not affect the expression of neurogenesis- and synaptic transmission-related genes in the olfactory bulb of European sea bass (Dicentrarchus labrax) ArchiMer
Mazurais, David; Servili, Arianna; Le Bayon, Nicolas; Gislard, Sebastien; Madec, Lauriane; Zambonino-infante, Jose-luis.
The decrease in ocean pH that results from the increased concentration of dissolved carbon dioxide (CO2) is likely to influence many physiological functions in organisms. It has been shown in different fish species that ocean acidification (OA) mainly affects sensory systems, including olfaction. Impairment of olfactory function may be due to a dysfunction of the GABAergic system and to an alteration of neuronal plasticity in the whole brain and particularly in olfactory bulbs. Recent studies revealed that OA-driven effects on sensory systems are partly mediated by the regulation of the expression of genes involved in neurotransmission and neuronal development. However, these studies were performed in fish exposed to acidified waters for short periods, of...

Moderate hypoxia but not warming conditions at larval stage induces adverse carry-over effects on hypoxia tolerance of European sea bass ( Dicentrarchus labrax ) juveniles ArchiMer
Cadiz, Laura; Ernande, Bruno; Quazuguel, Patrick; Servili, Arianna; Zambonino-infante, Jose-luis; Mazurais, David.
Environmental conditions, to which organisms are exposed during all their life, may cause possible adaptive responses with consequences in their subsequent life-history trajectory. The objective of this study was to investigate whether ecologically relevant combinations of hypoxia (40% and 100% air saturation) and temperature (15° and 20 °C), occurring during the larval period of European sea bass larvae (Dicentrarchus labrax), could have long-lasting impacts on the physiology of resulting juveniles. Hypoxic challenge tests were performed over one year to give an integrative evaluation of physiological performance. We revealed that juvenile performance was negatively impacted by hypoxia but not by the thermal conditions experienced at larval stage. This...

Digestive system functionality of fish larvae relies on the onset of genetically pre-programmed and extrinsically influenced digestive functions. This study explored how algal supplementation (green-water) until 14 days post hatch (dph) and the ingestion of food [enriched rotifer (Brachionus plicatilis) paste] from 15 dph onward affects molecular maturation and functionality of European eel larval ingestion and digestion mechanisms. For this, we linked larval biometrics to expression of genes relating to appetite [ghrelin (ghrl), cholecystokinin (cck)], food intake [proopiomelanocortin (pomc)], digestion [trypsin (try), triglyceride lipase (tgl), amylase (amyl)], energy metabolism [ATP synthase F0 subunit 6 (atp6), cytochrome-c-oxidase 1 (cox1)], growth...

Optimizing the metabolic use of these carbohydrate-rich plant products in fish feeds could be achieved through nutritional programming strategies of fish larvae. The present study aims at testing the long-term effect of an early-life high-carbohydrate nutritional (34% of starch in the feed) stimulus in European sea bass (Dicentrarchus labrax), by examining several biochemical and molecular parameters involved in carbohydrate metabolism in sea bass larvae and juveniles, compared to a control group fed standard larval regime (phase 1). As awaited, specific activities of digestive enzymes, from pancreatic and intestinal segments, involved in glucide digestion (amylase, sucrase) were enhanced together with an up-regulation of glucokinase gene and...

Regulation of the corticosteroid signalling system in rainbow trout HPI axis during confinement stress. ArchiMer
Kiilerich, Pia; Servili, Arianna; Peron, Sandrine; Valotaire, Claudiane; Goardon, Lionel; Leguen, Isabelle; Prunet, Patrick.
This study aims to shed light on corticosteroid regulation of stress in teleost fish with focus on the corticosteroid signalling system. The role of the mineralocorticoid-like hormone 11-deoxycorticosterone (DOC) in fish is still enigmatic, as is the function of the mineralocorticoid receptor, MR. Low plasma DOC levels and ubiquitous tissue distribution of MR question the physiological relevance of the mineralocorticoid-axis. Furthermore, the particular purpose of each of the three corticosteroid receptors in fish, the glucocorticoid receptors, GR1 and GR2, and the MR, is still largely unknown. Therefore we investigate the regulation of cortisol and DOC in plasma and mRNA levels of MR, GR1 and GR2 in the HPI-axis tissue (hypothalamus, pituitary and...

Salinity reduction benefits European eel larvae: Insights at the morphological and molecular level ArchiMer
Politis, Sebastian Nikitas; Mazurais, David; Servili, Arianna; Zambonino-infante, Jose-luis; Miest, Joanna J.; Tomkiewicz, Jonna; Butts, Ian A. E..
European eel (Anguilla anguilla) is a euryhaline species, that has adapted to cope with both, hyper- and hypo-osmotic environments. This study investigates the effect of salinity, from a morphological and molecular point of view on European eel larvae reared from 0 to 12 days post hatch (dph). Offspring reared in 36 practical salinity units (psu; control), were compared with larvae reared in six scenarios, where salinity was decreased on 0 or 3 dph and in rates of 1, 2 or 4 psu/day, towards iso-osmotic conditions. Results showed that several genes relating to osmoregulation (nkcc2 alpha, nkcc2 beta, aqp1dup, aqpe), stress response (hsp70, hsp90), and thyroid metabolism (thr alpha A, thr alpha B, thr beta B, diol, dio2, dio3) were differentially expressed...

Temperature is important for optimization of rearing conditions in aquaculture, especially during the critical early life history stages of fish. Here, we experimentally investigated the impact of temperature (16, 18, 20, 22 and 24°C) on thermally induced phenotypic variability, from larval hatch to first-feeding, and the linked expression of targeted genes [heat shock proteins (hsp), growth hormone (gh) and insulin-like growth factors (igf)] associated to larval performance of European eel, Anguilla anguilla. Temperature effects on larval morphology and gene expression were investigated throughout early larval development (in real time from 0 to 18 days post hatch) and at specific developmental stages (hatch, jaw/teeth formation, and first-feeding)....

Temperature induced variation in gene expression of thyroid hormone receptors and deiodinases of European eel ( Anguilla anguilla ) larvae ArchiMer
Politis, Sebastian Nikitas; Servili, Arianna; Mazurais, David; Zambonino-infante, Jose-luis; Miest, J. J.; Tomkiewicz, J.; Butts, I. A. E..
Thyroid hormones (THs) are key regulators of growth, development, and metabolism in vertebrates and influence early life development of fish. TH is produced in the thyroid gland (or thyroid follicles) mainly as T4 (thyroxine), which is metabolized to T3 (3,5,3’-triiodothyronine) and T2 (3,5-diiodothyronine) by deiodinase (DIO) enzymes in peripheral tissues. The action of these hormones is mostly exerted by binding to a specific nuclear thyroid hormone receptor (THR). In this study, we i) cloned and characterized thr sequences, ii) investigated the expression pattern of the different subtypes of thrs and dios, and iii) studied how temperature affects the expression of those genes in artificially produced early life history stages of European eel (Anguilla...

Ocean acidification is a recognized consequence of anthropogenic carbon dioxide (CO2) emission in the atmosphere. Despite its threat to marine ecosystems, little is presently known about the capacity for fish to respond efficiently to this acidification. In adult fish, acid-base regulatory capacities are believed to be relatively competent to respond to hypercapnic conditions. However, fish in early life stage could be particularly sensitive to environmental factors as organs and important physiological functions become progressively operational during this period. In this study, the response of European sea bass (Dicentrarchus labrax) larvae reared under three ocean acidification scenarios, i.e., control (present condition, P-CO2 = 590 mu atm, pH total =...

The Medio-Basal Hypothalamus as a Dynamic and Plastic Reproduction-Related Kisspeptin-gnrh-Pituitary Center in Fish ArchiMer
Zmora, Nilli; Stubblefield, John; Golan, Matan; Servili, Arianna; Levavi-sivan, Berta; Zohar, Yonathan.
Kisspeptin regulates reproductive events, including puberty and ovulation, primarily via GnRH neurons. Prolonged treatment of prepubertal striped bass females with kisspeptin (Kiss) 1 or Kiss2 peptides failed to enhance puberty but suggested a gnrh-independent pituitary control pathway. Kiss2 inhibited, but Kiss1 stimulated, FSh beta expression and gonadal development, although hypophysiotropic gnrh1 and gnrh receptor expression remained unchanged. In situ hybridization and immunohistochemistry on brains and pituitaries revealed a differential plasticity between the 2 kisspeptin neurons. The differences were most pronounced at the prespawning phase in 2 regions along the path of gnrh1 axons: the nucleus lateralis tuberis (NLT) and the neurohypophysis....
